MEXICO CITY - The Mexican navy said on Wednesday it had captured one of the leaders of the Zetas drug cartel, a notoriously brutal gang reported to be breaking apart due to an internal feud.

The navy said it had caught the man it believed to be Zetas boss Ivan Velazquez in the central state of San Luis Potosi, in a boost to outgoing President Felipe Calderon's efforts to crack down on the violent cartels.

The Zetas have perpetrated some of the most sickening acts of Mexico's drug war and continued to expand even as rival gangs joined forces against them. They are now regarded as one of the two most powerful drug cartels in the country.

Velazquez is due to be paraded before the media on Thursday morning as is customary with such captures in Mexico.

The suspected gang leader surrendered to the navy in the city of San Luis Potosi without a shot being fired, an eyewitness told Reuters.

Known as "Z-50" or "El Taliban," Velazquez has been one of the leading figures in the Zetas. Formed by a group of army deserters in the late 1990s, the gang acted as enforcers for the Gulf Cartel before splitting with their employers in 2010.

Longstanding rivalry between the Zetas' top leader, Heriberto Lazcano, and his second-in-command Miguel Trevino has exploded into violence, raising fears the hostilities could bring a fresh wave of bloodletting.

Switched allegiance
The Zetas boast 10,000-plus gunmen, and the prospect of them fighting for control of local trafficking networks and smuggling routes has alarmed security experts.

A video "mockumentary" that shows children as kidnappers, corrupt cops and drug traffickers sparked a fierce debate in violence-torn Mexico. Msnbc.com's Dara Brown reports.

Earlier this week, Mexican news magazine Proceso reported that Velazquez had switched his allegiance to the Gulf Cartel due to a rupture with Trevino, citing messages posted online.

Velazquez is listed by the government as one of the country's most-wanted drug kingpins. The Mexican government has offered a reward of up to 30 million pesos ($2.34 million) for information leading to his arrest.

Debate rages over Mexico 'spillover violence' in US

Since 2009, more than 20 drug lords have been caught or killed. The most recent capture came two weeks ago, when the navy arrested Gulf Cartel head Jorge Costilla, alias "El Coss."

Earlier on Wednesday, the navy announced the capture of 18 suspected Zetas in the northern state of Nuevo Leon.

Mexico's drug war: No sign of 'light at the end of the tunnel'

How to contain the threat posed by the drug gangs is one of the main challenges facing Calderon's successor Enrique Pena Nieto, who is due to take office on December 1.

About 60,000 people have been killed in drug-related violence during Calderon's six-year term.

Probing matters of the heart: Stem cell differentiation study sheds light on genetic basis of heart disease

ScienceDaily (Sep. 14, 2012) ? The fate of an embryonic stem cell, which has the potential to become any type of body cell, is determined by a complex interaction of genes, proteins that bind DNA, and molecules that modify those genes and proteins.

In a new paper, biologists from MIT and the University of California at San Francisco have outlined how those interactions direct the development of stem cells into mature heart cells. The study, the first to follow heart-cell differentiation over time in such detail, could help scientists better understand how particular mutations can lead to congenital heart defects. It could also assist efforts to engineer artificial heart tissue.

"We're hoping that some of the information we've been able to glean from our study will help us to approach a new understanding of heart development, and also lead to the possibility of using cells that are generated in a dish to replace heart cells that are lost as a consequence of aging and disease," says Laurie Boyer, an associate professor of biology at MIT and a senior author of the paper, which appears in the Sept. 13 online edition of Cell.

Research in Boyer's lab focuses on how DNA is organized and controlled in different cell types to create the wide variety of cells that make up the human body.

Inside a cell, DNA is wrapped around proteins called histones, which help control which genes are accessible at any given time. Histones may be tagged with different chemical modifications, which influence whether a particular stretch of DNA is exposed or hidden.

"These modifications cause structural changes that can act as docking sites for other factors to bind," says Joe Wamstad, a postdoc in Boyer's lab and one of the lead authors of the Cell paper. "It may make the DNA more or less accessible to manipulation by other factors, helping to ensure that you don't express a gene at the wrong time."

In this paper, the researchers found that histone-modification patterns shift rapidly as a stem cell differentiates. Furthermore, the patterns reveal genes that are active at different stages, as well as regulatory elements that control those genes.

Tracking development

To discover those patterns, the researchers grew mouse embryonic stem cells in a lab dish and treated them with proteins and growth factors that drive heart cell development. The cells could be followed through four distinct stages, from embryonic stem cells to fully differentiated cardiomyocytes (the cells that compose heart muscle). At each stage, the researchers used high-throughput sequencing technology to analyze histone modifications and determine which genes were being expressed.

"It's basically watching differentiation over time in a dish, and being able to take snapshots of that and put it all together to try to understand how the complex process of cardiac commitment is regulated," Boyer says.

The researchers found that they could identify groups of genes with related functions by comparing their modification patterns and whether they were being transcribed at a particular time. They also identified regulatory regions located far away from the genes they regulate. Many of these regions were located in sections of DNA previously thought to be "junk." Recent studies have revealed that much of this DNA actually plays important roles in regulating gene expression.

"We're starting to link genes with the regulatory elements that may be activating them, and beginning to draw a picture of the molecular circuitry that is controlling and driving these cardiac-specific programs -- the DNA elements that are important for turning on all the genes that you need to make a heart cell," Wamstad says.

The team also identified transcription factors -- proteins that initiate the expression of genes -- that appear to work collaboratively at regulatory regions to drive transcription of genes important for cardiac development. Defects in many of these transcription factors have previously been linked to congenital heart defects.

Linking disease with DNA

Previous genome-wide sequencing studies have revealed genetic variations that are more common in people with congenital heart disease or cardiovascular disease. The data obtained in this study should help researchers figure out why those variations give rise to such diseases.

The findings could also help researchers figure out how to create heart cells for possible therapeutic use, says Richard Lee, a professor of medicine at Harvard Medical School and member of the Harvard Stem Cell Institute.

"It's a roadmap of how the heart cell develops, and it's a tremendous achievement," says Lee, who was not part of the research team. "The work shows that the heart cell develops through coordinated gene expression transitions, and how those transitions are controlled. This study will help us to understand how to make new heart cells, which has been a big challenge for treating heart failure."

The researchers are now looking for other combinations of transcription factors that are involved in controlling cardiac differentiation. They are also studying variations in the regulatory sequences they identified to figure out how these sequences might give rise to congenital heart disease or susceptibility to cardiovascular disease later in life.

"I am all for pension and benefits reform when done in a proper and judicious manner that looks ahead at future retirees, whether in the public or private sector. However, it is a dangerous precedent to not fulfill the promise of pension and health benefits made to workers who entered the system while it was in existence," Samuels said.

"My father, Howard Samuels, was the first chairman of the NYC OTB under Mayor John Lindsay," Samuels continued, "and he would be appalled, as am I, that nearly 40 years later, the government is breaking its promise to these retirees by not continuing their health benefits when they are most needed."

More than a year ago, an appeals court affirmed the city should not be on the hook for funding the workers' health care.

A bill now online in the state Legislature, if passed, "Directs the state to reimburse the city of New York for the health care benefits of the retirees of the New York city off-track betting corporation; appropriates $7,000,000 therefor."

Cuomo vetoed a similar bill last year, and today, a Cuomo spokesman declined comment on his intentions, if any, regarding this year's reincarnation.

Bills, county agree: Extend lease by 1 year

ORCHARD PARK, N.Y. (AP) ? The Buffalo Bills and Erie County agreed in principle Wednesday to a one-year lease extension that would allow the team to keep playing at Ralph Wilson Stadium next season.

County executive Mark Poloncarz announced the agreement after Bills CEO Russ Brandon raised concerns about the lack of progress in negotiations to reach a long-term deal, and with the current lease set to expire July 31. Poloncarz said the agreement was reached after several discussions with team officials, including a conversation with Brandon on Monday.

"Nobody wants to have a gun at their heads thinking if we don't get it done by the end of the season then what's going to happen," Poloncarz said. "By doing an extension, it guarantees the Bills are here for the full 2013 season, and it gives us ample time during the next 18 months or so to continue these negotiations."

Poloncarz said the state is also on board with a one-year extension, and expects a formal deal to be signed "fairly quick."

The announcement came after Brandon went public by informing The Buffalo News that negotiations had stalled in a story published Wednesday morning. Brandon then repeated his concerns in an interview with The Associated Press.

"They have stalled over the last few months, and hopefully, we'll restart the process and reset the calendar in the near future," Brandon said. "We've been ready to go and have been in discussions with the appropriate parties for many months now. Over the last few months, we've hit somewhat of a cone of silence in the discussions."

The lack of progress has already led to one setback.

Brandon said the team will miss a deadline this month to apply for an NFL loan assistance program to help offset the $200-$220 million in costs the Bills and taxpayers would share for proposed renovations and upgrades to the 39-year-old stadium.

Brandon said the Bills needed to present their proposal for loan assistance at a committee meeting to have it placed on the agenda for a league meeting next month. The next chance the Bills will have to do that won't be until March.

However, a one-year extension fails to address a key concern in securing the franchise's long-term future in Buffalo. The team's founder and hall of fame owner Ralph Wilson is 93, and he spent a week in the hospital this month.

Wilson has said he has no intention to leave the team to his family, and instead plans to have his heirs sell the franchise. That opens the possibility of a new owner relocating the franchise, and makes a move less expensive if the team is not tied to a long-term lease.

"I'm not going to sit here and say that Mr. Wilson's age or health is not a factor because it is," Poloncarz said. "I'm not going to ignore it. ... No. We're going to continue to move forward. And I'm hopeful that we're going to get a lease done as soon as possible. I'm just disappointed it's not going to get done by the end of this year."

Poloncarz expects the new one-year deal would be similar to the team's existing lease.

Poloncarz and the state have a meeting set for Sept. 21, but noted the last time the three parties all met together was June 29.

Lt. Gov. Robert Duffy disputed claims that negotiations have stalled, saying state representatives have been in constant negotiations with the team and local government for the last several months.

"Let's set the record straight: the governor and this administration are actively negotiating in good faith with the Bills," Duffy said in a released statement.

"Keeping the Bills in Buffalo is critically important to the state and the region. And this administration is committed to doing everything we can to ensure that the Bills remain in western New York," Duffy said. "If taxpayers are going to be asked for substantial financial support for stadium improvements, they must be assured of a long term commitment by the Bills to stay in Buffalo regardless of future ownership."

Josh Vlasto, spokesman for New York Gov. Andrew Cuomo, noted that the state hired attorney Irwin Raij last month to assist in negotiations. Raij specializes in stadium development projects and lease agreements. He was most recently represented Guggenheim Baseball Management in buying the Los Angeles Dodgers.

Vlasto also noted that Cuomo included spending millions of dollar to retain the Bills among his list of "critical economic development projects."

The state has a large stake in keeping the Bills, because the team is estimated to generate between $15 million and $20 million in state taxes. Erie County also has a stake, because it controls the stadium lease.

Brandon sidestepped a question of how confident he was that a deal with all the parties would be completed.

"I'm very confident that we have our ducks in the proverbial row to sit down and have meaningful and thoughtful conversations with our stakeholders," he said. "It's our No. 1 initiative. So we're always ready and willing to talk at the drop of a hat. We're ready to go at any point, and have been over a year."

The Bills are also negotiating with Rogers Communications to renew a five-year series of home games Buffalo plays in Toronto. The current deal runs out this year after the Bills "host" the Seattle Seahawks in Canada's largest city and financial capital on Sept. 16.

Brandon said he expects to resume discussions with Rogers officials within the next three weeks. He said talks were delayed because the team was focused on training camp, the start of the season and lease negotiations.

Brandon said lease talks will have no effect on renewing the deal to play in Toronto.

Rodgers said.? ?It?s just frustrating when you?re positive that there?s either a missed call, or that the rule was not interpreted the way that it?s supposed to be interpreted.? There were multiple instances of that, and when you watch the film back it?s frustrating.

?That being said, there were just some bizarre calls on both sides.? Anybody who watches the TV copy, I mean I saw it from the sidelines, but we scored a touchdown on a legit block in the back.? I don?t know what happened on that.? It has to hopefully get better.?

Despite those concerns, the NFL has benefited from the reality that, based on the images pumped into TVs throughout the country, the officials look the part, act the part, and sound the part.? Brushing off mistakes by pointing out that the regular officials make mistakes, too, the league has crafted a simple but highly effective P.R. strategy that will keep fans from reacting in the only way that matters ? by not buying tickets and/or not watching games.

Even with high-profile players like Rodgers speaking out, only a major gaffe that directly influences the outcome of a game will even begin to move the needle.

We?re not saying it?s right, but it?s reality.? And so the locked-out officials can wait for a major blunder that prompts a fan outcry ? or the locked-out officials can get back to work.

Chemists develop nose-like array to 'smell' cancer

ScienceDaily (Sep. 13, 2012) ? In the fight against cancer, knowing the enemy's exact identity is crucial for diagnosis and treatment, especially in metastatic cancers, those that spread between organs and tissues. Now chemists led by Vincent Rotello at the University of Massachusetts Amherst have developed a rapid, sensitive way to detect microscopic levels of many different metastatic cell types in living tissue.

Findings appear in the current issue of the journal ACS Nano.

In a pre-clinical non-small-cell lung cancer metastasis model in mice developed by Frank Jirik and colleagues at the University of Calgary, Rotello's team at UMass Amherst use a sensor array system of gold nanoparticles and proteins to "smell" different cancer types in much the same way our noses identify and remember different odors. The new work builds on Rotello and colleagues' earlier development of a "chemical nose" array of nanoparticles and polymers able to differentiate between normal cells and cancerous ones.

Rotello explains, "With this tool, we can now actually detect and identify metastasized tumor cells in living animal tissue rapidly and effectively using the 'nose' strategy. We were the first group to use this approach in cells, which is relatively straightforward. Now we've done it in tissues and organs, which are very much more complex. With this advance, we're much closer to the promise of a general diagnostic test."

Until now the standard method for precisely identifying cancer cells used a biological receptor approach, a protein binding to a cancer cell wall. Its major drawback is that one must know the appropriate receptor beforehand. Rotello and colleagues' breakthrough is to use an array of gold nanoparticle sensors plus green fluorescent protein (GFP) that activates in response to patterns in the proteins found in cancer cells within minutes, assigning a unique signature to each cancer.

The chemist says, "Smell 'A' generates a pattern in the nose, a unique set of activated receptors, and these are different for every smell we encounter. Smell 'B' has a different pattern. Your brain will instantly recognize each, even if the only time you ever smelled it was 40 years ago. In the same way, we can tune or teach our nanoparticle array to recognize many healthy tissues, so it can immediately recognize something that's even a little bit 'off,' that is, very subtly different from normal. It's like a 'check engine' light, and assigns a different pattern to each 'wrong' tissue. The sensitivity is exquisite, and very powerful."

For this work, the researchers took healthy tissue and mouse tumor samples and trained the nanoparticle-GFP sensor array to recognize them and the GFP to fluoresce in the presence of metastatic tissue. Metastases are differentiated from healthy tissue in a matter of minutes, providing a rapid and very general means of detecting and identifying cancer and potentially other diseases using minimally invasive microbiopsies.

"It's sensitive to really subtle differences," says Rotello. "Even though two cheeses may look the same, our noses can tell a nicely ripe one from a cheese that's a few days past tasting good. In the same way, once we train the sensor array we can identify whether a tissue sample is healthy or not and what kind of cancer it is with very high accuracy. The sensitivity is impressive from a sample of only about 2,000 cells, a microbiopsy that's less invasive for patients."

In addition to the high sensitivity, the authors point out, their sensor is able to differentiate between low (parental) and high (bone, adrenal, and ovary) metastases, as well as between site-specific cells such as breast, liver, lung and prostate cancers.

"Overall, this array-based sensing strategy presents the prospect of unbiased phenotype screening of tissue states arising from genetic variations and differentiation state." Their next step will be to test the new sensor array method in human tissue samples, the researchers say.

Within an era of a growing reliance on digital technologies to instantly and effectively express our values, allegiances, and multi-faceted identities, the interest in digital research methodologies among Sociologists of Religion comes as no surprise (e.g. Bunt 2009; Cantoni and Zyga 2007; Contractor 2012 and Ostrowski 2006;Taylor 2003). However the methodological challenges associated with such research have been given significantly less attention. What are the epistemological underpinnings and rationale for the use ?digital? methodologies? What ethical dilemmas do sociologists face, including while protecting participants? interests in digital contexts that are often perceived as anonymised and therefore ?safe?? Implementing such ?digital? research also leads to practical challenges such as mismatched expectations of IT skills, limited access to specialized tools, project management and remote management of research processes.

Hosted by the Centre forSociety, Religion, and Belief at the University of Derby and funded by Digital Social Research, this conference will bring together scholars to critically evaluate the uses, impacts, challenges and future of Digital Methodologies in the Sociology of Religion. We envisage that the conference will lead to an edited textbook and are currently in discussion with key publishers. For the purpose of the conference and textbook, digital research is broadly defined as research that either works within digital contexts or which uses either online or offline digital tools. Abstracts for papers that focus on one, or more, of the following themes are invited:

(AP) ? Democrats open their national convention Tuesday offering President Barack Obama as America's best chance to revive the ragged U.S. economy and asking voters to be patient with incomplete results so far. Michelle Obama, in her opening-night speech, aims to give people a very personal reminder of "the man that he was before he was president."

"The truth is that he has grown so much, but in terms of his core character and value, that has not been changed at all," Mrs. Obama said.

Obama, campaigning in Norfolk, Va., predicted he'd get "all misty" watching his wife's speech from the White House with their two daughters.

"Whatever I say here today, it's going to be at best a distant second to the speech you will hear tonight from the star of the Obama family," he told a friendly crowd at Norfolk State University.

The three-day convention has drawn thousands of delegates to a state Obama narrowly carried in 2008. And although Obama no longer is the fresh-faced newbie who leveraged a short Senate career into an audacious run for the nation's highest office, he still can excite partisans, and Democrats were counting on massive numbers to pack an outdoor stadium for his speech later in the week ? weather permitting.

Virginia Sen. Mark Warner and a host of Democratic allies worked to rev up delegate enthusiasm, saying Obama has a strong record to defend. They noted the president had helped the economy rebound, presided over an increase in the stock market and brought troops home from Iraq and Afghanistan.

"We've got some truth telling to do," Warner told Florida delegates at a breakfast meeting. "America is better off today than it was four years ago when this president took over."

Newark, N.J., Mayor Cory Booker told the delegates Democrats need to get fully behind Obama, comparing the differences between a large voter turnout in his home state during the 2008 election and a more modest outpouring one year later, when his home state elected Republican Chris Christie as governor.

"Change is never made in a sedentary position," he said.

The Democrats dispatched U.S. Senate candidate Elizabeth Warren, who hopes to unseat Republican Scott Brown in Massachusetts, to make the case for Obama on morning talk shows, and she acknowledged that "it's tough out there" for many Americans.

New government reports out Tuesday underscored that reality.

U.S. factory activity shrank for the third straight month in August and construction spending fell in July from June by the largest amount in a year. Next up: The Labor Department will release new jobs numbers Friday. Economists expect to see some job gains but believe the unemployment rate will remain at 8.3 percent.

Obama told the Norfolk crowd that GOP nominee Mitt Romney and running mate Paul Ryan "want to take us backwards." He urged voters to stick with the Democrats to "close the gap between what America should be and what it is right now."

The president dropped off a case of White House-brewed beer at a Norfolk fire station before returning home, telling the firefighters, "I want you to test it out."

Ryan, the Wisconsin congressman, kept the GOP focus trained on the question of whether Americans are better off than they were four years ago, just as Ronald Reagan asked about Jimmy Carter's tenure decades earlier.

Voters "fired Carter and they hired Reagan, and we're going to do the same thing this time," Ryan told a cheering crowd in Westlake, Ohio.

Romney's campaign reinforced the economic message with a new Web video answering Obama's statement that "there are always going to be bumps on the road to recovery." The new video showcases a series of ordinary people who've lost their jobs saying, "I'm an American, not a bump in the road."

Romney, his convention behind him, drove from his vacation home in New Hampshire to the Vermont home of former Massachusetts Lt. Gov. Kerry Healey to spend the day preparing for the fall debates with Obama. His campaign raised at least $100 million last month, for the third consecutive month, The Associated Press has learned.

Romney raised more money than Obama in May, June and July, including money collected by the Democratic Party. Obama's advisers are now publicly acknowledging the president will likely be outspent.

Democrats released a party platform for ratification Tuesday that echoes Obama's call for higher taxes on the wealthy and reflects his shift on gay marriage by supporting it explicitly.

In a nod to dissenters on gay marriage, the platform expresses support for "the freedom of churches and religious entities to decide how to administer marriage as a religious sacrament without government interference."

As with the deeply conservative Republican platform, not all of which Romney endorses, nothing binds Obama to the specifics of the party's manifesto.

Michelle Obama, in an interview airing on SiriusXM's "The Joe Madison Show," said she wants to use her opening speech to "remind people about the values that drive my husband to do what he has done and what he is going to do for the next four years. I am going to take folks back to the man he was before he was president."

San Antonio Mayor Julian Castro delivers the convention's keynote address Tuesday night, a nod to the importance of Hispanic voters in the race.

"Under any score ? immigration, education, health care ? in any number of issues, he has been a very effective advocate for the Latino community," Castro said of Obama during an interview on CNN.

Obama campaign spokeswoman Jen Psaki said Castro's story "reflects the president's story and the American story" of working hard, playing by the rules and getting a fair shot in the land of opportunity. Castro "will talk about those values" and outline the choice for voters, she said.

With flourishes but no suspense, Democrats will march through the roll call of states re-nominating Obama for president and Joe Biden for vice president on Wednesday after former President Bill Clinton puts Obama's and Biden's names into nomination.

Obama's big acceptance speech is Thursday, and Democrats are closely monitoring the weather forecast since he's scheduled to speak outdoors at Bank of America stadium. Campaign aides said the speech would go on outdoors "rain or shine" unless the weather would put people at risk. The National Weather Service says there is a 40 percent chance of thunderstorms Thursday night:

The Democrats were working to leverage the impact of the convention: The first lady will present the Top 10 List on David Letterman's show Wednesday night. And, Obama told supporters by email that actor Kal Penn ? who worked in Obama's White House ? would host a special online program before Thursday night's convention speech featuring special guests and interviews with Mrs. Obama and Biden.

With polls showing a tight race, everything seemed to matter intensely.

In the swing state of Virginia, the state Board of Elections ruled Tuesday that conservative former Rep. Virgil Goode can appear on the state's presidential ballot. Republicans fear Goode will drain votes from Romney, and had tried to keep him off.

Democrats chose North Carolina for their convention to demonstrate their determination to contest it in the fall campaign. Obama carried North Carolina by 14,000 votes in 2008, but faces a tough challenge this time given statewide unemployment of 9.6 percent.
